Description

Transfected HEK293 cells were lysed via freeze-thaw cycles in ice-cold PBS with protease inhibitors. Lysates were clarified by 20, 000 g centrifugation (30 min, 4°C), then aliquoted, lyophilized, and stored at -80°C. Protein concentration was determined by BCA assay. Gene overexpression was confirmed by Western blot using anti-His tag and/or target-specific antibodies, with empty vector-transfected lysate as a negative control.
