Cell Death Caused by Iron Metabolism Disorder

Iron is in a dynamic balance of constant absorption, utilisation, storage and circulation in the body, known as iron homeostasis. This homeostasis is regulated by multiple metabolic pathways as well as enzymes and organelles, and acute and chronic cellular stress resulting from abnormal metabolic and biochemical processes may trigger widespread non-apoptotic forms of cell death.

  • Glutathione Depletion

Decreased glutathione peroxidase (GPX4) activity and reduced cellular antioxidant capacity lead to lipid peroxidation and metabolic dysfunction and increased lipid reactive oxygen species (ROS), which can trigger iron death.

Susceptibility to ferroptosis is closely linked to many biological processes, including amino acid, iron and polyunsaturated fatty acid metabolism, and the biosynthesis of glutathione, phospholipids, NADPH and coenzyme Q10, and is associated with pathological cell death associated with mammalian degenerative diseases, tumours, stroke, cerebral haemorrhage, traumatic brain injury, local ischaemia-reperfusion injury and renal failure.

Membrane Integrity Assay
