Cellular and Mitochondrial Iron Homeostasis

Cellular iron uptake is critical for meeting the cellular iron requirements of many metabolic processes, including electron transport, nucleotide synthesis, oxygen transfer, and redox reactions. The high redox potential of iron facilitates the reactions required for these processes, but it can also have pathological consequences, including increased mitochondrial reactive oxygen species (ROS) production and even cell death when iron homeostasis is disrupted. Therefore, precise cellular and mitochondrial iron uptake is essential for maintaining iron homeostasis and health.

Cellular Regulation of Iron

Iron metabolism is regulated at both systemic and cellular levels. At the cellular level, regulation of the expression of proteins involved in iron metabolism and homeostasis in vivo, such as ferritin or transferrin receptors, is coordinated by the interaction of iron-sensing proteins, called iron regulatory proteins (IRP) or IRE-binding proteins. Thus, the IRP/IRE system is a critical component of the organism, enabling cells to rapidly accommodate cytoplasmic iron and promote the function of many iron-dependent cellular components at the post-transcriptional level.

The intracellular iron pool is regulated by the binding of IRP1 and IRP2 to IRE. IRP1 and IRP2 are RNA binding proteins that interact with IRE to control the translation of proteins involved in iron metabolism. IRPs are either present at the 3′ UTR or the 5′ UTR of the target mRNA. When an IRP binds to a single IRE at the 5′ UTR, mRNA translation is repressed. On the other hand, the binding of IRP to IRE at the 3′ UTR stabilizes the transcript and leads to increased mRNA translation.
