Ferroptosis is defined as a form of iron-dependent regulated cell death caused by unrestricted lipid peroxidation and subsequent membrane damage, which can be triggered by extrinsic or intrinsic pathways.Ferroptosis can cause alterations in a wide range of physiological processes within cells, and Protheragen has experience in the field of cellular metabolism and provides our clients with imaging and assay services for Ferroptosis-related indicators and physiological processes.


Mechanism of Ferroptosis

Lipid peroxidation (the process of oxidative degradation of lipids by free radicals) is the main biochemical event that triggers ferroptosis. Therefore, dedicated enzymes to increase free radical production, fatty acid supply and lipid peroxidation are key to Ferroptosis.

  • Free Radical Generation
    by radicals, including reactive oxygen species (ROS) and reactive nitrogen species (RNS), which are produced by iron-dependent Fenton reactions, mitochondria, or enzymes of the nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (NOX) family. Excess ROS produced as a by-product of oxidative phosphorylation may cause oxidative damage to mitochondrial proteins, membranes, and DNA, thereby impairing mitochondrial function.
  • Fatty Acid Delivery
    Lipid droplets can release fatty acids under stressful conditions to amplify lipid peroxidation during ferroptosis by activating autophagy. A low-fat diet may be a safe and beneficial strategy to prevent ferroptosis-related diseases.
  • Lipid Peroxidation
    Lipid peroxidation is the most important cause of ferroptosis. Intracellular lipid oxidation metabolism is impaired and abnormal metabolism occurs under the catalytic effect of iron ions, and when the cellular antioxidant capacity is weakened, lipid reactive oxygen species accumulate, causing intracellular redox imbalance and inducing cell death.
