Iron Metabolism-related Gene Identification Service

Iron is essential for life, but it can also lead to cell death. Cells have therefore evolved a robust and tightly regulated set of genes to maintain iron homeostasis in animals. Protheragen offers gene identification services related to iron metabolism in animals to help our clients conduct in-depth research on related topics.

Regulatory Genes for Iron Metabolism

Iron homeostasis is the result of a combination of pathways and 4 major cell types: enterocytes, hepatocytes, macrophages, and erythroblasts. The EGF/EGF receptor signaling pathway, heme production, STAT signaling, cyclic AMP signaling, ferritin storage and bone morphogenetic protein-SMAD signaling are all involved in iron regulation. So far, genes related to the regulation of iron metabolism are still being discovered.

Intracellular Iron Metabolism Regulation

Intracellular iron metabolism is mainly controlled through the coordinated post-transcriptional regulation of various iron metabolism genes. Many of the mRNAs involved in iron metabolism contain

  • Iron Response Elements (IREs): IREs can bind two functionally similar iron regulatory proteins, IRP1 and IRP2. Depending on whether the IRE is located in the 5'-UTR or the 3'-UTR, the IRE-IRP interaction has an opposite effect on target gene expression.
  • ZRT/IRT-like Protein (ZIP) Family: The ZIP family also has an important role in iron transport, with ZIP14 mediating the uptake of non-transferrin-bound iron (NTBI) by directly transporting NTBI across the cell membrane.
  • The PCBP Family: Each member of the PCBP family contains three highly conserved K homology (KH) structural domains and can form complexes with other family members, capable of acting as cytosolic iron molecular chaperones to deliver iron to ferritin.

Our Services

Iron metabolism is regulated by a variety of genes and pathways, and many genes involved in the regulation of iron metabolism are still being discovered. With years of research in the field of biometabolism, Protheragen offers several different genetic screening strategies to assist our clients in their iron metabolism related research.

In particular, we also provide iron metabolism-related gene identification service in plants, and if you are interested in our services, please don't hesitate to contact us.

Genetic Identification Strategies

  • Screening with Libraries
Using the following libraries, we perform high-throughput screening (HTS) or phenotypic analysis using cellular signaling pathway reporters for the identification of genes related to iron metabolism.

Our Advantages

Chemical Compound Libraries

With our Drug Discovery and Target Identification services, we find chemical compounds modulate iron metabolism pathway or disease of interest and identify cellular targets of the compounds.

Our Advantages

shRNA or siRNA Libraries

Genome-wide or targeted RNAi screens are performed by gene knockout identification of genes associated with iron metabolism pathways or genes associated with mutant phenotypes. The program can target genes from multiple species including human, mouse, and rat.

Our Advantages

CRISPR sgRNA Libraries

We offer high-throughput manipulating gene expressions genome-widely or in specific pathways using CRISPR sgRNA libraries. We provide CRISPR KO (knockout), CRISPRi (interference) and CRISPRa (activation) screening, data analysis and target validation to ensure correct results.

  • Interacting Protein Analysis by Co-IP/MS
The coupling of mass spectrometry (MS) with co-immunoprecipitation (Co-IP) allows efficient identification of interacting proteins of the protein primarily targeted by antibody. We offer Co-IP/MS service to uncover interacting proteins of your protein of interest,as well as Protein Interaction assays to better confirm the results.
  • Comparative Transcriptomics
With our next generation sequence (NGS) platform, we offer transcriptome sequence and DE analysis to find genes and pathways associated with gene modification, drug treatment or other altered conditions in your samples.
  • In vitro Pulldown and MS
We offer in vitro pulldown services that use proteins, DNA oligomers, RNA oligomers, small molecule chemicals or other suitable biomolecules as bait to capture the proteins bound to them and thereby characterize the function of the target gene.
