Ferroptosis plays a crucial role in cell proliferation, senescence and differentiation. At the same time, iron levels in cells need to be strictly balanced, as excess iron can impair cell function and eventually lead to cell death due to ROS production. In addition, higher levels of iron transport proteins increase iron-mediated ROS, which subsequently leads to Ferroptosis. iron-mediated ROS is important for ferroptosis, and the production of ROS during iron metabolism mainly results from the "Fenton reaction" and Haber-Weiss reaction processes.

Ferroptosis Regulation

Ferroptosis can be triggered by structurally diverse small molecules or prevented by lipophilic antioxidants. Ferroptosis is caused by elevated levels of ROS due to elevated intracellular iron concentrations and depletion of the antioxidant GSH leading to lipid peroxidation, which leads to cell death.

  • GPX4
    GPX4 is an antioxidant enzyme that protects cells and membranes from peroxidation by using glutathione as a cofactor for GPX4 to neutralize lipid peroxidation and protect membrane fluidity. It is a specific and powerful central regulator of Ferroptosis when GPX4 is directly inhibited or indirectly inactivated by glutathione depletion.
  • GSH
    GPX4 acts as an antioxidant by catalyzing its substrate GSH, therefore, GSH indirectly affects GPX4 activity. Meanwhile, Buthionine sulfoximine (BSO) induces ferroptosis by inhibiting glutathione levels. BSO is an inhibitor of glutamate-cysteine ligase, the rate-limiting enzyme of glutathione synthesis.
  • NRF2
    NRF2 is a stress-inducible transcription factor whose protein levels are maintained at low levels by three different E3-ubiquitin ligase complexes. In disease settings with high NRF2 levels, such as cancer, tumor cells can use these protective detoxification systems to prevent lipid peroxide accumulation and lipid oxidation of target proteins to survive; whereas in diseases with low NRF2 levels, such as neurodegenerative diseases, increased lipid oxidation and inactivation of downstream NRF2 targets can significantly enhance overall protein lipid oxidation and Ferroptosis thereby promoting disease progression.

TBA
Malondialdehyde (MDA) is one of the lipid peroxidation products, which can interact with TBA (thiobarbituric acid) to produce a pink compound with a peak absorption at 532nm.

TBARS
TBARS can therefore be used as a better indicator of lipid peroxidation. The TBARS assay provides the ability to analyze levels of free radical activity in disease states and to detect the properties of many antioxidants.
