Adenoviral Vector

Adenoviruses are a class of medium-sized, non-enveloped, double-stranded DNA viruses that have proven to be highly valuable in research. Adenoviral vectors have several advantages, including their large packaging capacity, high titers, and efficient transduction of both dividing and non-dividing cell types. These vectors can deliver large amounts of genetic material to target cells, making them ideal for gene therapy applications. Unlike retroviruses, adenoviruses do not integrate into the host genome, preventing potential insertional mutagenesis. However, this also results in transient transgene expression, as the viral genome does not persist in the host cells.

Adenoviruses can be generated through homologous recombination of adenovirus plasmids, with the AdEasy™ system being the most widely used method. This system involves the recombination of shuttle vectors and adenoviral vectors, allowing for the efficient production of adenoviral particles. The transgene of interest is cloned into the shuttle vector, which is then recombined with the adenoviral plasmid. The resulting recombinant plasmid is verified, linearized, and transfected into packaging cells to produce adenovirus particles. Adenoviral vectors provide researchers with a powerful tool for delivering genetic material to a wide range of cell types, making them invaluable in various fields of research and potential clinical applications.
