Tet Regulatory Protein Expression Vector

The Tet regulatory protein expression vector is a versatile tool in molecular biology research that allows for the inducible and controlled expression of genes in mammalian cells. This vector builds upon the Tet System, which was initially developed by Gossen and Bujard in 1992 and involves the use of tetracycline or its analogs as inducers.

The Tet system relies on the interaction between the tetracycline repressor (tetR) and the tetracycline response element (TRE) to regulate gene expression. The TRE consists of seven repeats of a 19-nucleotide sequence that tetR recognizes. In the absence of tetracycline, tetR binds to the TRE, allowing for transcription and gene expression. However, in the presence of tetracycline, tetracycline molecules bind to tetR, preventing its interaction with the TRE and leading to reduced gene expression.
