Description
ALKBH8 is a protein consisting of 664 amino acids and is encoded by a gene located on chromosome 11. It belongs to the AlkB family of proteins and contains one RNA recognition motif (RRM) domain. ALKBH8 is a homolog of the Escherichia coli protein AlkB. The main function of ALKBH8 is to protect DNA and RNA from damage caused by methylating compounds present in the environment. It achieves this by directly reversing cytotoxic alkylation lesions, specifically 1-methyladenine (1-meA) and 3-methylcytosine (3-meC), in both DNA and RNA molecules. To carry out its enzymatic activity, ALKBH8 utilizes ferrous iron and alpha-ketoglutarate as cofactors, with 2-oxoglutarate serving as a co-substrate. Molecular oxygen acts as the oxidizing agent during oxidative demethylation. It's worth noting that there are three isoforms of ALKBH8 due to alternative splicing events occurring in the gene.
