Guidelines

1. Prior to resuspension, give the tube a quick spin in the centrifuge to make sure the freeze-dried siRNA settles at the bottom.
2. Stir the siRNA oligos into DEPC water until you reach the desired concentration. For instance, you can dissolve one vial of 5 nmol siRNA oligo in 250 µl of DEPC water to end up with a final concentration of 20 µM.
3. 48 to 72 hours before lysing the cells, transfect them with an amount of siRNA ranging from 10 nM to 100 nM.

Quality Control

The process of oligonucleotide synthesis is carefully tracked on a base by base basis using trityl analysis. This ensures the efficiency of the coupling process. After that, the oligonucleotide is purified by carrying out affinity-solid phase extraction. A mass spectrometer is used to further investigate the annealed RNA duplex, confirming the precise composition. Each batch is compared to the one before it using mass spectrometry to achieve the highest level of consistency between batches.
